When it comes to nicotinamide to treat cancer, there are more studies in human cells and in animals than in actual humans. In these few human trials, there is some evidence supporting the use of oral nicotinamide for preventing the formation of skin cancer in at-risk individuals.

In terms of actual cancer treatment, clinical trials suggest that nicotinamide may improve the efficacy of radiotherapy for tumor control and quality of life for some patients, particularly those with laryngeal or bladder cancer. However, not all of the study results were favorable, as some patients experienced nausea and vomiting with no therapeutic benefit. Other notable findings, such as re-established sensitivity of resistant breast-cancer cells to chemotherapy, have yet to be replicated in human trials. More studies are needed on nicotinamide treatments for cancer.

The review doesn’t provide enough information to determine a dosage range, but mentions that oral administration of nicotinamide is safe and well tolerated for doses below 3 g/day.
